Production of Superoxide Radical by Reaction of Phenazine Methosulfate with Erythrocyte Membranes

Abstract
In aerobic reaction mixture containing phenazine methosulfate(PMS) and nitroblue tetrazolium(NBT), the addition of erythrocyte membrane fraction caused the reduction of NBT. The NBT reduction was inhibited almost completely by superoxide dismutase and partially by catalase, indicating that superoxide radical and hydrogen peroxide are produced in this reaction. The reduced NAD(NADH) was not required for this reaction to occure.
When phosphatidylcholine from egg yolk in place of erythrocyte membrane was added to the PMS+NBT reaction mixture, the reduction of NBT occurred more markedly than in the case of erythrocyte membrane addition, and it was also inhibited markedly by superoxide dismutase.
These results suggest that the production of superoxide radical by reaction mixtures containing erythrocyte membrane, PMS and NBT is due to the reaction of PMS with phosphatidylcholine component in the erythrocyte membranes.
